While you can do the basics of supervising on a mission you don't know, it is better if you know the basic outline of the missions. That helps  you know how to help the crew on the bridge as well as create interesting second stories at 2nd Chair.
Here you'll find a list of mission scripts from the Atlantis and Columbia. It is important to remember that sometimes missions change; they may not be the exact same as the script due to flight director styles and different paths the crews choose to take.
